Fluorescent plasmid transformation was applied to engineer biocontrol Bacillus strains. Combined with laser scanning confocal microscopy and root microbial community sequencing, the whole process of pathogen infection and antagonistic bacterial colonization was monitored.
Key Findings:
An in vivo fluorescent visualization method was established for real-time monitoring of tripartite microbial interactions in the banana rhizosphere.
Bacillus strains are recruited to the apical root infection sites by banana root exudates, suppressing the spread of Fusarium via niche competition and secretion of antimicrobial metabolites.
This work provides an in-situ visualization technique to optimize the application timing and dosage of biocontrol agents in field practice.

Summary:A fluorescently labelled strain system was constructed to track the tripartite interactions among Bacillus spp., Fusarium oxysporum f. sp. cubense Tropical Race 4 (Foc TR4) and banana plants in vivo, so as to dissect the root colonization pattern of biocontrol strains and the inhibition mechanism of banana Fusarium wilt.
